[Summary]The aim of this study is to obtain the gene expression profiles of the liver of young growing rats after mild restriction of food intake for one month.
[Overall design] Male Wistar rats, at 5 weeks of age, were fed with reduced diet (30% restriction) for one month (n=5). After 16h fasting, the liver was then extracted and total RNA was isolated using an RNeasy mini kit with DNase treatment (QIAGEN). The RNA samples were pooled from five rats for each group. Then, DNA microarray analysis was performed using duplicate GeneChips (Affymetrix) on each pooled RNA sample.

The expression profile of liver of rats fed Protein-restricted diet for 1 week

Male Wistar rats consumed a 12 g/100g casein diet between 10:00 and 18:00h for 3 d before they were given experimental diets. Three experimental diets, a 12 g/100 g casein diet and a protein-free diet were prepared(n=5). The experimental diets were given for 7 d on the same schedule.Total RNA was prepared from livers and pooled by each group. cRNAs from each group were used for hybridization to a rat Genome U34A Array (Affymetrix, Santa Clara, CA) according to the manufacture's protocol.
